Garlic (Allium sativum L.) cryopreservation by vitrification of apices

Abstract

A research has been developed at INIFAT’s Gene Bank for garlic in vitro conservation whit the purpose of recovering and preserving its variability. As a part of this research, this paper offers the results of the cryopreservation of clove apices. Vitrification with PSV3 solution (50% glycerol + 50% sucrose), during 3 hours, was the cryopreservation protocol. It was observed that a high quantity of apices (98.4%) continue growing after cryopreservation. Tree months after vitrification, 68.3% of apices developed plantlets that completed their development, and produced bulblets in a frequency similar than the control treatment. Hyperhydricity effect on plant development was evaluated. Four months after cryopreservation, 85.7% of plants evolved to bulblets of diameter from 2 to 5 mm. Plants have maintained the growth and viability. This fact offers a favorable perspective to bulblets diameter increase.
